Standard reference procedure for Neisseria gonorrhoeae culture and identification. |

Daily; preliminary report as soon as found positive, final for negative results at 3 days. |

Any site or fluid, urethral swab, endocervical swab, rectal swab or throat swab. |
| Collect: |
Inoculate Jembec plate of Modified Thayer Martin agar sealed in a plastic bag with a Co2 generating tablet.
Culture tests:
Test specific transport devices, recommended sources, transport temperature and requirements are required for each test method which include:
a. Culture,GC Screen vaginal, urethra, rectal, throat, any source.
1.) For culture of N. gonorrhoeae, use calcium alginate or dacron swabs for specimen collection. Cotton fibers contain fatty acids which are inhibitory to the gonococcus. Avoid swabs with wooden sticks.
2.) Inoculate culture directly onto a Jembec agar plate. Direct plating is necessary because the gonococcus is extremely susceptible to drying. Use one plate for each culture obtained. Roll the swab in a Z pattern across the agar surface.
3.) Label the plate with patient identification and specimen site.
4.) Place a CO2 generating tablet in the designated location in the Jembec plate and place in a plastic bag. Seal the bag.
5.) Transport immediately at ambient temperature. If there is any delay in transport, incubate at 37°C.
6.) For male patients, also submit a slide of urethral material for Gram stain. Gram stains of specimens from the endocervix are not as reliable. Stool is not an acceptable specimen for gonorrheal culture.
7.) If disseminated gonococcal infection is suspected, culture blood and suspicious sites such as petechiae or joint fluid. |

Culture for N. gonorrhoeae is the method of choice in cases of treatment failure and sexual abuse and for non-genital sources. Media and probes are available from laboratory or Regional Pathology Services. Unacceptable conditions: Delayed transport of the plates in non-CO2 environment. Refrigerate or frozen specimens. |
